Zhibai Dihuang Wan, a Chinese patent medicine. It is composed of Anemarrhena, Phellodendron, Rehmannia glutinosa, Cornus officinalis (processed), Peony Root, Chinese Yam, Poria, and Alisma. It has the effect of nourishing yin and reducing fire. It is used for yin deficiency and fire, hot flashes and night sweats, dry mouth and sore throat, tinnitus and spermatorrhea, and short and red urine.

Identification
1. Take this product and observe it under a microscope: the starch granules are triangular, oval or rectangular, with a diameter of 24-40 μm, and the umbilicus is short, slit-shaped or herringbone-shaped (Chinese yam). The irregular branched masses are colorless and dissolve in chloral hydrate test solution; the hyphae are colorless or light brown, with a diameter of 4-6 μm (Poria). The parenchyma is gray-brown to black-brown, and the cells are mostly wrinkled, containing brown nuclei (Rehmannia glutinosa). Calcium oxalate needles are bundled or scattered, 26-110 μm long (Anemarrhena asphodeloides). Calcium oxalate clusters exist in colorless thin-walled cells, and sometimes several are arranged in rows (Paeonia suffruticosa). The epidermal cells of the fruit peel are orange-yellow, and the surface view is polygonal, and the anticlimax is thickened in a beaded manner (Cornus officinalis). The thin-walled cells are round, with elliptical pits and integrated pit groups; the anticlimax of the endodermal cells is wavy, thick, lignified, and has sparse fine pores (Alismatis orientalis). The fiber bundles are bright yellow, and the surrounding cells contain prismatic calcium oxalate to form crystal fibers. The walls of the crystal cells are lignified and thickened (Phellodendron chinense).
2. Take 6g of honey pills of this product and grind them; or take 9g of small or large honey pills and cut them into pieces. Add 15ml of ether, shake for 15 minutes, let stand for 1 hour, filter, evaporate the ether from the filtrate, add 1ml of acetone to the residue to dissolve it, and use it as the test solution. Take another paeonol reference substance and add acetone to make a solution containing 1mg per 1ml, as the reference solution. According to the thin layer chromatography method (General Rule 0502), take 101 of each of the above two solutions and spot them on the same silica gel G thin layer plate to make strips, use cyclohexane-ethyl acetate (3:1) as the developing agent, develop, take out, dry, spray with hydrochloric acid acid 5% ferric chloride ethanol solution, and heat until the spots are clearly colored. In the chromatogram of the test product, the same blue-brown stripes appear at the corresponding position of the chromatogram of the reference substance.
3. Take 2g of this product, chop it, add 5ml of methanol, heat and reflux for 15 minutes, filter, take the filtrate, add methanol to make it 5ml, as the test solution. Take 0.1g of Phellodendron chinense reference medicinal material and make the reference medicinal material solution in the same way. Take berberine hydrochloride reference substance and add methanol to make a solution containing 0.5mg per 1ml as the reference solution. According to the thin layer chromatography method (General Rule 0502), take 1μl of each of the above three solutions and spot them on the same silica gel G thin layer plate, use toluene-isopropanol ethyl acetate methanol concentrated ammonia test solution (12:3:6:3:1) as the developing agent, place it in a developing cylinder pre-saturated with ammonia vapor, develop, take out, dry, and examine under ultraviolet light (365nm). In the chromatogram of the test product, the same yellow fluorescent spot appears at the corresponding position of the chromatogram of the reference medicinal material and the chromatogram of the reference substance.

Content determination
1. Dogwood
(1) Determined by high performance liquid chromatography (General Rule 0512).
(2) Chromatographic conditions and system suitability test: Octadecylsilane bonded silica gel was used as the filler; tetrahydrofuran-methanol-acetonitrile-0.05% phosphoric acid solution (1:4:8:87) was used as the mobile phase; the column temperature was 40°C; the detection wavelength was 236 nm. The theoretical plate number calculated based on the loganin peak should be no less than 4000.
(3) Preparation of reference solution: Take an appropriate amount of loganin reference substance, accurately weigh it, and add 50% methanol to make a solution containing 40% per 1 ml to obtain the test solution. Take the honey pills of this product, grind them, take 0.7 g, and accurately weigh it; or take the small honey pills or the large honey pills under the weight difference item, cut them into pieces, mix them, and take about 1 g and accurately weigh it. Place in a stoppered conical flask, accurately add 25 ml of 50% methanol, seal, weigh, ultrasonically treat (power 250 W, frequency 33 kHz) for 15 minutes, heat under reflux for 1 hour, cool, weigh again, make up the lost weight with 50% methanol, shake well, filter, accurately measure 10 ml of the filtrate, add to a neutral alumina column (100-200 mesh, 4 g, inner diameter 1 cm), elute with 50 ml of 40% methanol, collect the effluent and the eluate, evaporate to dryness, dissolve the residue with 50% methanol, transfer to a 5m volumetric flask, add 50% methanol to the scale, shake well, filter, take the filtrate, and obtain.
(4) Determination method: Accurately pipette 10 μl of the reference solution and the test solution respectively, inject into the liquid chromatograph, and determine.
(5) This product contains Cornus officinalis in the form of loganin (C17H26O10 ) . The content of each gram of water-honeyed pills shall not be less than 0.53 mg; each gram of small honeyed pills shall not be less than 0.40 mg; and each gram of large honeyed pills shall not be less than 3.6 mg .
2. Peony bark:
(1) Determined by high performance liquid chromatography (General Rule 0512).
(2) Chromatographic conditions and system suitability test: Octadecylsilane bonded silica gel was used as the filler; methanol and water (70:30) was used as the mobile phase; the detection wavelength was 274 nm, and the theoretical plate number calculated based on the paeonol peak should be no less than 3500.
(3) Preparation of reference solution: Take an appropriate amount of paeonol reference substance, weigh it accurately, and add methanol to make a solution containing 15 μg per 1 ml.
(4) Preparation of test solution: Take the honeyed pills of this product, grind them, take 0.4g, and weigh accurately; or take the small honeyed pills or the large honeyed pills under the weight difference item, cut them into pieces, mix them, take 0.5g, and weigh accurately. Place them in a stoppered conical flask, accurately add 50ml of 50% methanol, seal it tightly, weigh it, ultrasonically treat it (power 250W, frequency 33kHz) for 45 minutes, let it cool, weigh it again, make up the lost weight with 50% methanol, shake it well, filter it, and take the filtrate to obtain the solution.
(5) Determination method: Accurately pipette 10 μl of the reference solution and 20 μl of the test solution respectively, inject into the liquid chromatograph, and determine.
(6) This product contains peony bark in the form of paeonol (C 9 H 1 0 O 3 ). The content of each gram of water-honeyed pills shall not be less than 0.80 mg; each gram of small honeyed pills shall not be less than 0.55 mg; and each gram of large honeyed pills shall not be less than 5.0 mg.
